Polyclonal Antigen Preparation FAQ
 
 
1) In what form should I provide my antigen?
The antigen can be in a liquid (buffer), lyophilized, or in a gel slice.

2) Can a GST fusion protein be used?
Yes, any fusion protein can be used for the immunization phase, but we will need the cleaved protein for screening or another GST fusion protein to use as a negative control.

3) Can the protein be attached to beads?
Yes, for the immunization phase, but we will need purified protein to use in the ELISA screening phase.

4) How should I ship the antigen?
We recommend that you send it by overnight delivery on dry ice or ice packs.

5) Do you offer peptide synthesis?
Yes, we use an outside source. The cost is $44.00 per amino acid for 20 mg of peptide at >80% purity (determined by mass spec/HPLC). This takes an additional 2-3 weeks.

6) Does my peptide need conjugation?
If your peptide is less than 15 KD (molecular weight), we strongly recommend conjugation; if your peptide is greater than 25 KD, conjugation is generally not necessary. Anything in between is at your discretion. Usually, we conjugate 5 mg of the peptide to KLH for immunizations.

7) When can you start my project?
We can start the project within 48 hours after receipt of immunogen, provided we have a PO#.

8) What buffer components can I use with my antigen?
We cannot accept immunogens in buffers that have any of the following conditions (for immunizations):

  • Contain ANY metal dyes
  • Contain KCl, NaCl, or MgCl2 at > 1.0 M
  • 5% SDS (w/o consulting our veterinarian)
  • 6M Urea (w/o consulting our veterinarian)
  • 1% b-octylglucoside, a detergent
  • 1% Triton 100, a detergent
  • 1% Tween 20, a detergent
  • > 20% glycerol
  • 0.1mM PMSF (Protease Enzyme inhibitor)
  • 1ug/ml Leupeptin (Protease Enzyme inhibitor)
  • 1ug/ml Pepstatin A (Protease Enzyme inhibitor)
  • 0.1mM DIPF (Protease Enzyme inhibitor)
  • 3M DTT (a reducing agent)
  • 3M imidizole
